表达CGRP的HSV-I型扩增子载体对大鼠蛛网膜下腔出血迟发性血管痉挛保护作用的研究 被引量:1

摘要 目的探讨表达CGRP的HSV-I型扩增子载体对大鼠蛛网膜下腔出血迟发性血管痉挛的保护作用。方法枕大池二次注血法建立大鼠SAH后CVS动物模型,取脑,用HE染色法和TUNEL染色法检测对照组、缺氧缺血组和重组载体治疗组脑组织中凋亡细胞数量和脑组织神经病理改变。结果缺氧缺血后脑组织内的凋亡细胞明显增加,神经细胞变性明显。治疗组标记的阳性细胞数比同时间的缺氧缺血组明显降低,神经细胞变性坏死减少。结论重组载体对大鼠缺氧缺血性脑损伤有保抗作用,其作用与抑制细胞凋亡有关。 Objective To investigate the neuroprotective effect of neurotropic herpes simplex virus type i amplicon vector hearing CGRP gene on Cerebral Vasospasm (CVS) Model after Subarachnoid Hemorrhage in Rats.Methods Brain vascular vessel,smooth muscle and nerve celia enhancement, necrosis and apoptosis alteration were detected through HE and TUNEL. Results It is found that the numbers of apoptosis cell were markedly increased in hypoxic-ischemic brain tissue and decreased significantly in treated group compared with that in hypoxic-ischemic group. The neuronal degenerations in cortex were reduced in treated group. Conclusion The data indicate that neurotropic herpes simplex virus type i amplicon vector besting CGRP gene has protective effect on CVS, which has relation with inhibiting apoptosis.
