摘要
目的建立ELISA测定血清瘦素的方法。方法制备瘦素单克隆抗体,以双抗体夹心法测定血清瘦素。结果方法的平均回收率为89.7-92.1%,取浓度为1.21μg/L、6.32μg/L瘦素标本其批内变异分别为4.4%和2.6%;批间变异分别为5.7%和4.9%。方法的线性为:1.25-20μg/L。与放射免疫法比较有良好的相关性(r2=0.9988)。参考值范围为:5.26-5.92μg/L。结论该法简便、快速、特异、敏感,适于临床常规应用。
Objective To establish a ELISA for measuring serum leptin.Methods Preparation of monoclonal antibody for the examination of serum hptin by ＂sandwich＂ method.Results The analytical recovery were 89.7 - 92.1% at 1.25 - 20 gg/L of lep- tin. The within-assay were 4.4 % and 2.6 % at 1.21 gg/L and 6.32 gg/L of leptin. The between-assay were 5.7 % and 4.9% at 1. 21/q/L and 6.32 gg/L of hptin.The li~.afity of method was 1.25- 20 gg/L.There was good correlation between ELTSA and RIA （ r2 -- 0.9988）. The range of reference value were 5.26 - 5.92 pg/L. Conclusion The method is simple, rapid, precise and sensi- tive. ？ It＇s suitable for clinical routine determanifion.
