摘要
目的 建立毛细管电泳电导法测定广地龙中琥珀酸含量的方法。方法 以1.0mmoL/L Tris-10mmoL/L H3BP3-0.5%四乙烯五胺-0.025mmol/L十六烷基三甲基溴化铵为分离介质,使用未涂层融硅毛细管柱(50cm×75μm),分离电压-10kV,电导检测器检测。结果 琥珀酸的线性范围5.00~80.0μg/mL(r=0.9998),平均加样回收率98.4%,RSD=2.2%。结论 该法操作简便,快速。
OBJECTIVE A capillary electrophoresis with conductivity detection method to determine succinic acid in Pheretima aspergillum was described. METHODS A fused-silica capillary tube(50cm×75 μm)was used, at the separation potential of -10kV succinic acid was detected directly in running medium of 1.0 mmol/L riffs 10 mmol/L H3 BO3-0.5% tetren-0. 025 mmol/L CTAB( pH = 11.45). RESULTS The linear range of succinic acid was 5.00μg/mL - 80.0μg/mL( r =0.999 8), the average recovery of added sample was 98.4% with the RSD = 2.2%. CONCLUSION This method is simple and quick.
