摘要
目的探讨同源盒gax基因在破裂腹主动脉瘤组织中的表达水平及其分子生物学机制.方法破裂腹主动脉瘤体14个,破口位于前壁者4个,后壁者10个,正常腹主动脉组织(自愿捐肾者3人)3块.即刻切取破缘或正常血管组织冻存,分别将其设为实验组(A组)和对照组(B组).采用逆转录多聚酶链反应(RT-PCR)和蛋白印迹杂交(Western blot)检测二组组织中gax的表达.结果gax基因的mRNA与蛋白表达态:在A组分别是(144.9±38.7)与(75.8±12.3);在B组中分别是(99.4±30.7)与(46.8±18.7),组间比较P<0.05.结论同源盒gax基因可能是参与腹主动脉瘤病理演变后期过程中比较重要的基因之一.
Objective To Investigate the molecular biology mechanism and express level of homeobox gax gene in the human abdominal aortic aneurysm (AAA) rupture. Methods Specimens were collected from 14 patients with the ruptured (group A) AAA and from 3 healthy volunteers for the donated kidney (group B), and studied in order to assess the level of gax gene expression by means of a reverse transcription - polymerase chain reaction ( RT - PCR) and Western bolt analysis. Results There was remarkable difference in the expression of gax gene between with A group (144. 9 ±38.7 vs 75.8 ± 12. 3) and B group (99. 4 ±30. 7 vs 46. 8 ± 18.7) at a mRNA and protein level ( P 〈 0. 05 ) . The expression ratio of gax gene was increased nearly double in group A compared with group B. Conclusions These results suggest that homeobox gax gene might be involved in a pa thological change of human rupture AAA, and play a significant role in the late process of the aneurysm rupture.
