固定化细胞联产L-苹果酸和L-天冬氨酸的研究 被引量:3

摘要 用延胡索酸铵为底物溶液,通过串联的固定化延胡索酸酶产生菌细胞和固定化L-天冬氨酸酶产生菌细胞的两柱式反应器,延胡索酸定量地被转化为L-苹果酸和L-天冬氨酸。转化液用浓盐酸调至pH 2.78,L-天冬氨酸沉淀析出,过滤洗涤后可得L-天冬氨酸产品。滤液中加入氢氧化钙,使苹果酸转为苹果酸钙沉淀,再经酸化、离交、浓缩、结晶,可得L-苹果酸产品。使用该法,可有效地降低L-苹果酸产品中延胡索酸含量,简化工艺,提高底物延胡索酸的利用率。 A new method for united production of L-malic acid and L-aspartic acid by immobilized Brevibacteriun flavum 865 cells producing fumarase and immobilized E. coli 8906 cells producing aspartase was investigated. The substrate of ammonium fumarate was first passed through the column bioreactor containing immobilized fumarase and converted into L-malic acid with 70% yield. Then, the unconverted substrate was passed through another column bioreactor containing immobilized aspartase and converted into L-aspartic acid with almost 100% yield. The simplicity of the procedure, the full utilization of fumarate and the low content of fumarate in L-malic acid product were the advantages of this new method.
