摘要
采用荧光素染色法,以ACAS570型粘附细胞仪研究ATP敏感性钾通道特异性激动剂cromakalim对正常心肌细胞内Ca2+的作用及对缺氧再给氧时细胞内Ca2+的影响。结果显示:cro-makalim降低正常心肌细胞内Ca2+的浓度;同时明显减轻缺氧再给氧时细胞内Ca2+超载现象。二者均呈良好的量效关系。提示:cromakalim可能有减轻缺氧再给氧损伤的作用。
Byusingfluorescencestainingmethod,theefectofcromakalimonintracelularfrecalcium(Ca2+)levelinculturedneonatalratmyocardialcelsandinanoxia-reoxygenationmyocardialcelswerestudiedwithadherentcelanalysisandsortingcytometer570(ACAS570).Theresultsdemonstratedthat(1)ThelevelofintracelularCa2+decreasedimmediatelyaftertheadditionofcro-makalim;(2)CromakalimsignificantlyreducedthelevelofintracelularCa2+duetoanoxia-reoxy-genation.Aloftheseweredose-dependent.Theseshowedthatcromakalimcouldagainstcalciumoverloadduringanoxia-reoxygenation.
