超负荷心房肌肌球蛋白基因表达的研究 被引量:3

摘要 为了研究心肌肌球蛋白基因表达在心房超负荷中的作用,我们用斑点杂交的方法,测定了20例心脏病患者右心房心肌肌球蛋白重链(MHC)的mRNA表达。男11例,女9例,平均年龄31.7岁(5~52岁)。结果显示:超负荷的心房肌MHCmRNA表达异常,表现为α-MHC下降,β-MHC升高;且右心房腔扩大愈明显,β-MHC愈高,呈显著正相关。提示:α-MHC向β-MHC基因转化与心房重塑程度相关。由于β-MHC为胚胎表型,易于衰竭,可能促进心房重塑。 Forevaluatingtheroleofcardiacmyosinheavychain(MHC)geneexpressioninover-loadinghumanatrium,therelativeamountofMHCisoforminrightatrialmyocardiumsuferingfromheartdiseasewasinvestigatedusingdot-blotmethods.Elevenmals,ninefemaleswereincluded,withameanageof31.7years(5-52years).TheresultsshowedthatMHCmRNAexpressionwasabnormalinoverloadingrightatriaum:alpha-MHCmRNAexpressiondecreased;beta-MHCmRNAincreased.Rightatrialandrightventriclesizeweresignificantlypositivelycorrelatedwithbeta-MHCmRNAexpression,indicatingthatthegeneswitchingfromalpha-MHCtobeta-MHCcorrelatedwiththedegreeofatriumandventricleremodeling.Beta-MHCisafetalphenotype,thereforemayleadtotheappearanceofmyocardialcelsthat“wearout”atanacceleratedrate,therebycontributingtotheprogreessionofatriumdilatationandremodeling.
