摘要
研究了外源激素对不同小麦品种成熟种子体细胞胚发生的影响。结果表明:2,4-D明显促进小麦胚性愈伤组织的发生,其诱导效应表现出基因型间的差异。诱导体胚发生的最适宜浓度为2,4-D 2.0~4.0 mg·L^1,附加KT对胚状体的萌发有显著的促进作用。另外,及时调整培养基,适当的降低激素含量对体胚的诱导也非常重要。晋农211在2,4- D2.0 mg·L^1的MS培养基上,45 d后转移至2,4-D浓度1.0 mg·L^1+KT0.5 mg·L^1的培养基上胚状体诱导率较高。农大981胚性愈伤组织在2,4-D2.0 mg·L^1的MS培养基上,50 d降低2,4-D浓度至1.0mg·L^1+KT0.5 mg·L^1胚状体诱导率较高。
Effect of two exogenous hormones on frequency induction of wheat (Triticum aestivum L. ) somatic embryos is studied. The following results are revealed: Concentration of 2,4-D in media is necessary for induction of embryogenic, with a different frequency between the different genotype. The most appropriate concentration of 2,4-D is at 2. 0-4. 0 mg·L^-1This capability of induction embryos can be enforced by the addition of KT in medium. In addition, adjusting the medium in time and reducing the concentration of exogenous hormones is also a significant factor for the induction of embryos.
