氯嘧磺隆胶体金免疫检测试纸条的制备与应用 被引量:4

摘要 利用胶体金标记的经亲和层析纯化的氯嘧磺隆单克隆抗体,氯嘧磺隆-牛血清白蛋白,羊抗鼠IgG制备了氯嘧磺隆胶体金免疫检测试纸条。该试纸条的最低检测极限为100μg/L,检测时间10min。8个土壤样品分别添加0~2000μg/L的氯嘧磺隆标样,用0.15mol/L NaHCO,提取2h后用气相色谱和试纸条进行检测,如果以100μg/L作为阳性与阴性样品的区分标准,气相色谱和试纸条检测结果完全一致。所制备的胶体金免疫检测试纸条可以用于土壤中氯嘧磺隆残留的筛选。 A monoclonal antibody-based lateral flow dipstick is developed in which the detection reagent is colloidal gold labeled affinity purifed monoclonal anti- chlorimuron-ethyl antibody. The capture reagent is chlorimuron-ethyl-bovine serum albumin conjugate and goat anti-mouse antibody which is immobilized on the nitric cellulose (NC) membrance. The visual detection limit for chlorimuron-ethyl is 100 μg/L. Aanalysis can be completed in less than 10 min. Eight soil samples were fortified with chlorimuron-ethyl in a concentration range of 0 -2000 μg/L and extracted with 0.15 mol/L NaHCO3 for 2 h at room temperature. The supernatant was collected and analyzed directly with dipstick test and gas chromatography. When the cut-off value was set at 100 μg/L to distinguish the positive and negative samples, The results from the two methods are completely identical. The monoclonal antibody-based lateral flow dipstick can be employed for rapid and simple screening for the residue of chlorimuron-ethyl in soil samples.
