摘要 目的:研究黄苓茎叶活性部位(SLSBGAF)的抗病毒作用。方法:体外细胞培养观察1:2~1:256不同稀释度SLSBGAF对柯萨奇B族病毒(CoxB3、CoxB4、Cox B5)、流感病毒、副流感病毒、腺病毒(Adv3、Adv7)、呼吸道合胞病毒(RSV).疱疹病毒(HSV-1、HSV-2)等10种病毒致细胞病变(CPE)的抑制作用;流感病毒感染小鼠模型,分别观察SLSBGAF对小鼠肺指数、间接荧光染色观察肺内病毒增殖的影响,并以小鼠存活时间和死亡率为指标观察药物的保护作用。结果:SISBGAF对CoxB3等10种病毒所致细胞病变有明显的抑制作用,有效浓度为0.20~1.56mg/mL,IC50值为0.08~0.34mg/mL,治疗指数(TI)值分别为4和8;sLSBGAF(54.3,108.5,217mg/kg)灌胃给药对流感病毒引起的小鼠肺炎有治疗作用,肺指数和病毒的肺内增殖低于对照组,并呈一定的量效相关性;此外对流感病毒所致小鼠死亡有不同程度的保护作用,217mg/kg剂量组作用明显,死亡保护率为40%,生命延长率为39.08%。结论:SLSBGAF可用于流感病毒、副流感病毒、柯萨奇病毒等相关感染性疾病的预防和治疗。 Aim: To investigate the antiviral effect of the active fraction from stems and leaves of Scutellaria baicalensis Georgi. (SLSBGAF). MethodS:To observe the inhibiting effect of cytopathic effect (CPE) of SLSBGAF, ten kinds of viruses, such as Coxsachie virus ( Cox B3, Cox B4, Cox B5 ), influenza virus, parainfluenza virus, adenovirus ( Adv3, Adv7), respiratory syncytial virus(RSV) and herpes simplex virus (HSV-1, HSV-2) were cultured in different dilution from 1 : 2 to 1 : 256. Lungs of mouse model infected by influenza virus were observed to estimate the impact of SLSBGAF on the lung index and virus reproduction in lung which was observed by indirect fluorescence staining. Mouse survival time and death ratio were measured to evaluate the protective effect of SISBGAF. Results : SLSBGAF has significant inhibiting cytopathic effect (CPE) on the ten virues. The effective concentration varied from 0. 20 mg/ mL to 1.56 mg/mL, IC50 varied from 0.08 mg/mL to 0. 34 mg/mL, TI calculated at 4 and 8; Intragastric administration with SLSBGAF of 54.3 mg/kg, 108.5 mg/kg, and 217 mg/kg has therapeutic efficacy on mouse pneumonia from influenza virus, lung index and virus reproduction in lung were lower than those of control group. There even existed a certain degree of dose-response relationship. Administration of SLSBGAF at the different dose also resulted in different degrees of protective effects on mouse death suffered from influenza virus, and dose of 217 mg/kg has led to the death protective ratio was 40%, the rate of life prolong was 39.08%. Conclusion: SISBGAF could be worth further investigation in the prevention and treatment of influenza virus, parainfluenza virus, Coxsachie virus-infected diseases.
