摘要
目的探讨水蛭提取液对实验性脑血肿周围组织热休克蛋白70(HSPTO)及转移生长因子β-1(TGFβ-1)表达的影响。方法采用成组设计的随机对照研究,用定量胶原酶注人大鼠尾状核建立脑出血模型,并于制模后随机分为水蛭提取液治疗组和生理盐水对照组,分别经尾静脉注射水蛭提取液和生理盐水,每日1次;制模后每日给予神经功能缺损评分,术后第3d、6d及10d通过免疫组化染色观察鼠脑血肿周围脑组织HSPTO与TGFβ-1的表达,图像分析仪计算阳性细胞面积。结果脑出血后两组神经功能缺失评分均随时间延长而显著改善(P〈0.01~0.001),治疗组评分各时间点明显优于对照组(均P〈0.01);术后第3d、6d及10d治疗组脑血肿周围组织HSPTO及TGFβ—1阳性细胞面积显著高于对照组(均P〈0.05)。结论水蛭提取液对实验性脑出血血肿周围组织保护作用机制可能与HSPTO与TGFβ—1表达增强有关。
Objective To study the effects of hirudo extract liquor (HEL) on the expressions of HSP70 and TGFβ-1 in intracerebral perihematoma tissues of rats. Methods We established the experimental ICH models in Wistar rats by stereotaxical injecting quantitative collagenase (0. 7 U collagenaseⅦ ) into their left caudate nuclei. The rats continued to be treated with HEL (treatment group) or normal saline (control group) through intravenous injection by vena caudalis and the scores of neurologic impairment in two groups were evaluated every day. The slices of these samples at 3rd, 6th and lOth day were stained by immunohistochemistry and the positive cells of HSP70 and TGFβ-1 were counted by image analysis system, respectively. Results The scores of neurologic impairment in two groups were remarkably reduced with time going(P 〈0. 01 ~0. 001 ). In addition,each time point, the recovery of neurological function in the treatment group was much better compared with control group ( all P 〈0.01 ). Moreover, at 3rd, 6th and lOth day, HSPTO and TGFβ-1 positive cell areas in treatment group were obviously higher than those in control group (all P 〈 0. 05 ). Conclusion HEL may promote the expressions of HSP70 and TGFβ-1 to protect intracerebral perihematoma tissues of rats.
