摘要
目的探讨转录因子E2F1基因的表达与胃癌生物学行为及生存素(Survivin)和P53表达的相关性。方法应用免疫组织化学(SP法)检测61例胃癌组织中E2F1、Survivin和P53的表达,并以20例胃低级别上皮内瘤变为对照。结果①E2F1、Sur-vivin和P53在胃癌组织中的阳性率分别为75.4%、85.2%和68.9%,显著高于低级别上皮内瘤变组织中的30.0%、40.0%和15.0%,有显著差异(P<0.05)。②E2F1的表达在胃癌组织分化程度、浸润深度、TNM分期淋巴结转移、远处转移中差异无显著性(P>0.05)。Survivin和P53在胃癌有无远处转移和TMN分期之间差异有显著性(P<0.05),且Survivin在有无淋巴结转移间差异有显著性(P<0.05)。③在胃癌中E2F1的表达强度分别与Survivin和P53的表达强度呈等级正相关(rs1=0.519,P1<0.01;rs2=0.602,P2<0.01)。结论E2F1可能作用于胃癌发生的早期阶段,Survivin和P53可能作用于胃癌发生的晚期阶段,三者在胃癌的发生、发展中可能起协同作用。
Objective To study the relationship between E2F1 gene expression and its biological behavior, as well as its correlation with the expression of Survivin, P53 in gastric carcinoma tissues. Methods The expressions of E2F1, Survivin and P53 were detected by immunohistochemical method (SP) in 61 gastric carcinoma tissues, and compared with those in 20 cases of low - grade intraepithelial neoplasia(IN)tissues. Results ( 1 ) The positive expression rates of E2F1, Survivin and P53 were 75.4%, 85.2%, 68.9% respectively in gastric carcinoma tissues, which were much higher than those (30.0%, 40.0% and 15.0% ) in IN tissues. No significant difference in E2F1 expression was found in different levels of differentiation grade, invasion depth, TNM stage, lymph node metastasis and distant metastasis (P 〉 0.05). (2)The expression of Survivin and P53 was higher in gastric carcinoma tissues compared with IN tissues in different levels of TNM classification and distant metastasis ( P 〈 0.05) and survivin expression was significantly different between non-and lymph node metastasis. (3)The expression of E2F1 was positively correlated with that of Survivin and P53 ( rs1 = 0.519, P1 〈 0.01 ; rs2 = 0. 602, P2 〈 0.01 ). Condusion E2F1 may play a role in the early phase, while Survivin and P53 may work in the late phase in carcinogenesis of gastric cacinoma.
