摘要 目的:研究HPV16型E7病毒癌基因感染对人宫颈癌HeLa细胞周期调节因子cdc25A mRNA表达的影响,探讨E7癌基因和cdc25A在宫颈癌发生发展中的作用。方法:用含有HPV16E7病毒癌基因的重组腺病毒载体感染体外培养的人宫颈癌HeLa细胞。采用反转录聚合酶链反应(RT-PCR)检测感染前后HeLa细胞cdc25A mRNA表达的差异。结果:感染后HeLa细胞cdc25A的mRNA含量较感染前显著增加(感染前灰度值0.2306±0.0964,感染后0.8739±0.2188,P<0.01)。结论:HPV16E7癌基因促进细胞周期调节因子cdc25A的表达,可能是宫颈癌发病机制中的重要分子事件。 Objective: To investigate the transcription of cdc25 A in HeLa cells after the transfection of human papillomavirus16 E7 oncogene by recombinant adenovirus vector, and to study the role of HPV E7 and cdc25A in the development of human cervical carcinoma. Method:As HeLa cells were transfected by recombinant adenovirus which expressing human papillomavirus 16 E7 gene ,reverse transcription polymerase chain reaction(RT-PCR) techniques were used to evaluate the measurements of cdc25A mRNA in HeLa cells. Result: RT-PCR measurements showed that the expression of cdc25A mRNA was increased significantly after transfeetion ( the gray numerical value : 0. 8739 ± 0.2188 vs 0.2306 ± 0. 0964, P 〈 0.01 ). Condusion: HPV16E7 oncogene enhanced the transcription of cdc25A ,which maybe an important event in the mechanism of the development of the cervical carcinoma.
