新抑癌基因ARHI在子宫内膜异位症中的表达及其意义 被引量:5

摘要 目的:研究子宫内膜异位症组织中抑癌基因ARHI蛋白的表达。方法:选择抗ARHI单克隆抗体,采用W estern blot免疫印迹法,对10例子宫内膜异位症的异位囊肿组织及其在位内膜组织进行蛋白检测,并以10例正常女性的在位内膜组织作对照。用Array 3000扫描仪扫描X线片,观察W estern blot的结果,用GDS8000型凝胶成像分析系统分析图片的辉度值和比值。结果:(1)ARHI在内异症异位内膜的表达高于在位内膜(P<0.001)及正常内膜(P<0.001);(2)内异症的在位内膜组与正常对照组比较,ARHI的表达无明显差异(P>0.05)。结论:抑癌基因ARHI在子宫内膜组织中普遍表达,且在异位内膜中表达上调,提示ARHI基因可能与子宫内膜异位症的发病及进展相关。 Objective:To investigate ARHI expression in endometriosis, and discuss the invasion mechanism of endometriosis. Methods:Ten specimens of ectopic and eutopic endometrium with endometriosis, and ten control members were investingated by using western blot. Array 3000 scanning apparatus scaned the X-ray films. The gray scale and the ratio were detected and analysed gy GDS8000 gal image analysis system. Results: ( 1 ) The expression of ARHI protein in ectopic endometrium group was extremely higher than that in eutopic group (P 〈 0.001 ) and that in normal endomerium group (P 〈0.001 ) ;(2) The expression of ARHI protein in eutopic endometrium group was not different with that in eutopic group ( P 〉 0.05 ). Conclusion: The tumor suppressor gene ARHI is generally expressed in endometrium, and especially higher in ectopic endometrium with endometriosis. ARHI may be involved in the development of endometriosis.
