槲皮素对H_2O_2所致乳鼠培养心肌细胞损伤的保护作用 被引量:5

摘要 目的观察槲皮素(QUE)对H2O2所致乳鼠培养心肌细胞损伤的保护作用。方法原代培养的新生Wistar乳鼠,随机分为正常对照组、H2O2损伤组和3种剂量QUE保护组。用比色法观察乳鼠心肌细胞培养液的乳酸脱氢酶(LDH)、脂质过氧化产物-丙二醛(MDA)的含量及超氧化物歧化酶(SOD)活性,并用电镜观察心肌细胞形态学改变。结果H2O2损伤组乳鼠心肌细胞培养液中LDH和MDA含量显著增高,SOD活性降低(与对照组相比,P<0.01),心脏超微结构损伤严重;QUE保护组与H2O2损伤组相比,LDH和MDA明显降低,SOD活性升高,心肌形态学变化减轻。结论槲皮素对H2O2诱导的乳鼠心肌细胞损伤有保护作用,其机制可能与清除氧自由基、抗脂质过氧化损伤有关。 Objective To investigate the protective effects of quercetin(QUE) on hydroperoxide-induced myocardial injury in neonate rats. Methods The primary cultured cardiomyocytes of neonate Wistar rats were randomly divided into 5 groups:normal control group, H2O2 induced myocardial injury group, and QUE partreatment groups ( with 10, 50 and 100μmol/L QUE). The lactate dehydmgenase (LDH) level, malondialdehyde (MDA) content and superoxide dismutase (SOD) activity of myocardial cell culture fluid in separate groups were detected. Changes of the cellular morphology were observed by electronic micmscope. Results The LDH level and MDA contents in myocardial cell culture fluid were significantly increased, the SOD activity was notably decreased,and the cardiac ultrastrueture injury was serious in H2O2 group,compared with control group. However, the QUE pretreatment groups with different dosage could prevented the alteration of the above-mentioned indexes. Conclusion QUE could prevent the myocardial injury induced by H2O2. Its possible mechamism might be concerned with scavenging oxyradical and anti-lipid peroxidation.
