摘要
目的研究不同剂量60Co射线照射后Fas、Egr-1在Hela细胞中的表达及其与细胞凋亡的关系;阐述其在放射诱导细胞凋亡中的作用。方法采用TUNEL法检测宫颈癌Hela细胞凋亡,半定量RT-PCR法检测Hela细胞Fas、Egr-1的表达。结果①照射后Hela细胞凋亡指数与对照组比较均有极显著差异(P<0.01);②照射后Hela细胞Fas表达与对照组比较均有极显著差异性(P<0.01);③照射后Hela细胞Egr-1表达与对照组比较均有极显著差异(P<0.01);④Fas表达与凋亡指数相关(r=0.947,P<0.01);⑤Egr-1表达与凋亡指数相关(r=0.969,P<0.01);⑥Fas表达与Egr-1表达相关(r=0.98,P<0.01)。结论①放射线可诱导Hela细胞中Fas、Egr-1的表达,并能诱导细胞凋亡;②Fas、Egr-1与细胞凋亡关系密切,二者呈正相关;③Egr-1可能通过上调Fas在Hela细胞中的表达而参与了放射诱导的细胞凋亡过程。
Objective To study the relationship between apoptosis and the expression of Fas and Egr-1 in cervical Hela cells that are subjected to different dosage of Co rays irradiation. To further investigate expression the role of Fas and Egr-1 radiation-induced apoptosis. Methods TUNEL technique was used to observe the apoptotic cervical carcinoma Hela cells. The expression of Fas and Egr-1 were detected by semi-quantitative RT-PCR. Results (1)Tne apoptosis ratios increased with dose ^60Co rays; (2)The mRNA expression of Fas genes in Hela cell were significantly higher than that in control ( P 〈 0.01 ) ; (3)Tne mRNA expression of Egr-1 genes in Hela cell were significantly higher than that in control (P 〈 0.01); (4)The mRNA expression of Fas genes was significantly correlated with the apoptosis ratios( r = 0. 947, P 〈 0.01 ) ; (5)The mRNA expression of Egr-1 genes was significantly correlated with the apoptosis rations ( r = 0. 969, P 〈 0.01 ) ; (6)The mRNA expression of Fas genes was significantly correlated with the mRNA expression of Egr-1 genes ( r = 0.98, P 〈 0.01 ). Condusion (1)radioactive ray can induce the expression of Fas and Egr-1 in Hela cell and it can induce apoptosis;(2)Fas and Egr-1 has a close relation to apoptosis and the twogenes are postitive correlated; (3)Egr-1 may regulate the expression of Fas in Hela cells to induce apoptosis.
