放疗联合斑蝥酸钠对肺癌A549细胞VEGF表达的影响

Influence of radiotherapy combined disodinm cantharidinate on the vascular endothelial growth factor expression in lung cancer A549 cell

目的探讨放疗联合艾易舒（斑蝥酸钠维生素B6注射液）对肺腺癌A549细胞放疗后VEGF表达的影响。方法MTT法观察艾易舒对A549细胞生长的影响，MTT法绘制A549细胞的细胞存活曲线，ELISA法检测细胞上清液中VEGF的浓度．结果放疗联合艾易舒可降低肺癌A549细胞放疗后VEGF的表达，并随浓度的增高而增强。单纯放疗后VEGF的平均值为73．50±1．74，艾易舒浓度为0．2mg／L、0．4mg／L、0．8mg／L时VEGF的平均值分别为58．88±1．33、54．37±1．72、39．75±1．08与单纯放疗后VEGF的平均值相比，0．4、0．8mg／L时P〈0．05．结论高、中剂量斑蝥酸钠联合放疗可降低肺癌A549细胞放疗后VEGF的表达。

Objective To study the influence of radiotherapy combined disodium cantharidinate on the vascular endothelial growth factor of lung adenocarcinoma cell A549 after radiotherapy. Methods The influence of growth on A549 cell was analysed by 3-（4,5-dimethytthiazole-2-yl）-2,5-diphenyl tetrazolium bromide （MTT） assay. MTT assay was drawn the A549 cell survival curve, and enzyme-labeled immunosorbent assay （ELISA） was detected the concentration of VEGF in the cell supernate. Results Radiotherapy combined disodium cantharidinate could reduce the VEGF expression of A549 cell after radiotherapy, and this effect was enhanced with the increasing concentration of disodium cantharidinate. The mean value of VEGF with simple postradiotherapy was 73.50±1.74, the concentration of disodium cantharidinate were 0.2, 0.4, 0.8mg/L, and the mean value of VEGF were 58.88±1.33, 54.37±1.72, 39.75±1.08, respectively. Compared to simple postradiotherapy, the concentration of disodium cantharidinate was 0.4 and 0.8mg/L （P〈0.05）. Condusions Middle and high dose of disodium cantharidinate combined radiotherapy can reduce the VEGF expression of A549 cell after radiotherapy.
