家蚕微孢子虫(镇江株)β-微管蛋白基因部分片段的克隆及系统发育分析 被引量:5

摘要 设计1对正、反向引物btubf/btubr,对家蚕微孢子虫(镇江株)基因组DNA的β-微管蛋白(beta-tubulin)基因进行扩增,得到部分片段。经PCR鉴定、酶切及测序分析,该片段与Nosema属其它微孢子虫的beta-tubulin同源。采用邻近归并法(Neighbour-Joining)构建系统发育树,结果表明微孢子虫和真菌关系密切。研究结果对于微孢子虫的系统发育研究和家蚕微粒子病的治疗具有积极意义。 A beta-tubulin DNA fragment of the Zhenjiang strain of Nosema bombycis was obtained by primers btubf/btubr. The amplified fragment was analyzed by PCR, restriction endonuclease digestion, and sequenced, which has high homology with other microsporidian beta-tubulins. Neighbour-Joining was used to construct phylogenetic tree, and it was verified that there was close relationships between microsporidian and fungi. The result has a positive significance on microsporidian phylogenetic research and microsporidiosis treatment.
