摘要 目的探讨固体硝酸盐还原酶试剂用于结核分枝杆菌药物敏感性试验的可行性。方法以标准绝对浓度法为参考方法,对79株不同耐药模式的结核菌进行固体硝酸盐还原酶药敏试验,并对两种试验结果进行kappa检验。结果三种一线抗结核药物的敏感性和特异性分别为:利福平:94.6%和97.6%;异烟肼:97.7%和88.2%;链霉素:97.2%和95.3%。经kappa检验,k系数分别为0.92,0.87,0.92。74.7%的菌株在7天就获得药敏试验结果,10天为98.7%,14天达100%。结论固体硝酸盐还原酶试剂配制简单,保存时间长,用于结核分枝杆菌药敏试验,省时且经济适用。 Objective To explore the feasibility of crystalline nitrate reductase reagent used in the drug susceptibility test of Mycobacterium tuberculosis. Methods Comparing with the standard absolute concentration method, 79 Mycobacterium tuberculosis strains with various resistance patterns were used in nitrate reductase drug susceptibility test, the consistency of the two methods was checked. Results Sensitivity and specificity of the two drugs were 94.6% and 97. 6% for rifampin, 97. 7% and 88. 2% for isoniazid, 97. 2% and 95.5% for streptomycin. Consistency check showed that k coefficient of the three drugs was 0.92, 0.87, and 0. 92. Results were available in 7 days for 74, 7 % of the strains, in 10 days for 98, 7 %, and in 14 days for 100 %. Conclusion Crystalline nitrate reductase reagent can be easily obtained and preserved for a long time, when used in the drug susceptibility test of Mycobacterium tuberculosis,time is shortened and of lower cost.
