摘要
目的:研究分离纯化的透明质酸寡糖片段(o-HA)对血管内皮细胞增殖的作用。方法:选用猪髂总动脉内皮细胞(PIEC)做为靶细胞,人胚肾293细胞做为对照细胞,应用本实验室分离纯化的o-HA(4-12-mer)配成一系列浓度(0、1、3、5、10及20μg/ml),加入靶细胞及对照细胞继续培养不同时间(0、6、12、24、48、72及96 h),从细胞计数、细胞周期分布和单层血管内皮细胞损伤修复来观察血管内皮细胞的增殖情况。结果:10μg/ml o-HA作用于PIEC时能显著促进增殖,继续增加o-HA的浓度,增殖作用未随之增强;o-HA刺激培养12 h PIEC即明显增生,至72 h达高峰(P<0.05);o-HA对PIEC损伤模型也有显著的促愈合作用。结论:o-HA能显著促进血管内皮细胞的增殖。
Objective:To investigate the effect of purified hyaluronan oligosaccharides (o-HA) on endothelial cells (EC) proliferation and wound healing. Methods:The purified hyaluronan oligomers below 6 disaccharide units (4-12- mer) were pooled and the biological effects of the mixture on EC proliferation and wound healing were studied. The cultured pig lilac endothelium cell lines (PIEC) added with o-HA (0, 1, 3, 5, 10 and 20 μg/ml) were observed at h 0, 6, 12, 24, 48, 72 and 96 respectively. The cell proliferation was determined by cell counting and flow cytometry. The effect of o-HA on PIEC wound healing was assayed in a wound healing model in vitro. Results; o HA at the concentration of 10 μg/ml significantly increased the proliferation of PIEC, but increasing the concentration of o HA did not show addi tional improvement of proliferation. Proliferation of PIEC occurred after o-HA treatment for 12 h, reached the peak at 72 h. At the same time, o-HA significantly enhanced the wound healing of PIEC at the concentration of 10 μg/ml. Conclusion: o-HA can promote wound healing and proliferation of EC in vitro.
