摘要
目的研究乳腺癌及其良性病变组织中MCP-1mRNA,MIP-1αmRNA表达并探讨其临床病理意义。方法40例乳腺癌和30例乳腺良性病例手术切除标本常规石蜡包埋切片,MCP-1mRNA和MIP-1αmRNA染色方法均为原位杂交法。结果40例乳腺癌MCP-1mRNA、MIP-1αmRNA阳性病例分别为25例(62.5%)和24例(60.0%),明显高于30例乳腺癌良性病例(均为4例,13.3%),均有高度显著差异(MCP-1:χ2=17.08,P<0.01,MIP-1α:χ2=12.56,P<0.01)。组织学分级Ⅰ+Ⅱ级、肿块最大径≤2cm和区域淋巴结未转移乳腺癌MCP-1mRNA,MIP-1αmRNA表达阳性率明显低于组织分级Ⅲ级、肿块最大径>2cm和区域淋巴结转移病例,均有显著或高度显著差异(P<0.05或P<0.01)。两者表达与乳腺癌患者年龄、月经状况、ER状态等临床特征无明显关系。两者在乳腺癌组织中表达阳性率之间存在密切正向关系(χ2=11.1,P<0.05)。结论MCP-1mRNA和MIP-1αmRNA表达可能是反映乳腺癌发生、进展及预后的重要生物学标志物,且两者可能具有相互协调作用。
Objective To study on the expressions of MCP - 1 mRNA, MIP - 1 αmRNA and detect their clinicopathological significances in the benigr and malignant lesions of marnmary gland. Methods In situ hybridization was used for the detecting expressions of MCP - 1mRNA and MIP - 1αmRNA on the routinely paraffin - embedded sections of the specimen of the benign and malignant lesions of mammary gland. Results The positive rate of MCP - 1mRNA, MIP - 1αmRNA was significantly higher in the breast cancer than that in the benign lesion of mammary gland (62.5 % vs 13.3 %, χ^2 = 17.05, P 〈 0.01; 60.0% vs 13.3%, χ^2 = 12.56, P 〈 0.01). The positive rates of MCP - 1 mRNA, MIP - 1 αmRNA were significantly lower in the cases of histologic grade Ⅰ + Ⅱ, maxial diameter of mass ≤ 2cm and without - metastasis of lymphnode than that in the ones of histological grade Ⅲ, maxial diameter of mass 〉 2cm and with - metastasis of lymphnode in the breast cancer (P 〈 0.05 or P 〈 0.01). No relationships were found among the expression of MCP - 1 mRNA and MIP - 1 αmRNA and other clinicopatholeo - gical features of breast cancer (P 〉 0.05). The closely positive correlation was found between the expression of MCP - 1mRNA and MIP - 1αmRNA in breast cancer (P 〈 0.01). Conclusions The expressicn of MCP - 1mRNA or MIP - 1 αmRNA might be important biological marker for the reflecting the carcinogenesis, progress, metastatic potential and prognosis of brest cancer, and two chemotaetic factor might have co - effect.
