摘要
以灭活大肠杆菌(E.coli)O157为抗原免疫产蛋鸡获得抗E.coli O157卵黄抗体(IgY),建立了该抗体的间接酶联免疫吸附试验(ELISA)检测方法。并研究了包被温度和时间及不同卵黄前处理方法对ELISA检测的影响,所获得的最佳检测条件为:以浓度为10^8~10^9/mL的灭活E.coliO157,为抗原进行包被,4℃包被19h,水稀释法对卵黄进行前处理,然后检测其中的IgY效价。该方法简便可靠,适于对大肠杆菌特异性抗体IgY的动态检测和批量检测。
Chicken was immunized with E. coli O157 as antigen,which produced a large amount of immunoglobulin of high titer in egg yolk,also called IgY. Indirect ELISA detection of IgY specific on E. coli O157 was studied. The parameters were as follows:polystyrene chloride microtitration plates were coated with E. coli O157 antigen at the amount of 10^8-10^9/mL,19 h at 4 C. The working concentration of HRP-labelled rabbit antichicken IgG was 1 : 320. Egg yolk pretreated with water-dilution improved the performance of ELISA detection. The ELISA system is dependable and good for the assay and surveillance of E. coli O157 specific IgY.
