摘要
在大鼠晶状体器官培养的条件下,运用单细胞电泳法(SCG),对远在晶状体混浊之前的晶状体上皮细胞进行了有关TNT致其DNA损伤(SSB)与修复的初步观察,提示DNA损伤也是体外TNT性白内障中的早期变化.
There is some restriction in the study on the effect of trinitrotoluene (TNT) on eye lens by using the model in vivo. Under the condition of organ culture, the model of TNT-cataract,because it takes the researchers longer time to establish the model of TNT-cataract was established,and using the single cell gel assay (SCG)technique was used to detect lens epithelial cells DNA single strand breaks(SSB) induced by TNT and its repair during the period long before opacity formation. Results showed that in DVA SSB lens epithelial cells occurred significantly at about 45 min after incubation with TNT (6. 5×10 ̄(-5) mol/L),but the morphological changes of lens seemed to appear after about 15 days. Incubating with different concentrations of TNT,the degree of DNA damage correlated positively with TNT concentration. Meanwhile,DNA repair also correlated positively with repairing time. The results suggested that DNA damage may be one of the most early important events during TNT cataractogenesis.
