A part of adhesive fibroblast in a fetal pancreatic cell culture was eliminated after 15h culturing. The cells that hadn't adhered were transferred and cultured in a fresh medium which contained 10% BS for two weeks, then the medium was replaced with a medium contained only 3% BS and was changed every week. The medium of the enriched pancreatic cells was used for isolation of insulin by CM-cellurose chromatography and sephadex G-75 gel filtration. The activity of insulin was measured by radioimmunoassay, and the results showed that the medium of the enriched pancreatic cell contained 286.7μU/ml of insulin and the specific activity of the partially purified insulin preparation was 473.4 μU/mg. When 0.2 mg(95μU)of this insulin was injected to mice(ip),the blood sugar content decreased about 30.7% within 30 min.
