大黄提取片对高脂饮食诱导肥胖大鼠脂代谢及相关基因表达的影响 被引量:18

Effects of Refined Rhubarb on Lipid Metabolism and Related Genes Expression in High Fat Diet Induced Obese Rat

摘要 目的:观察大黄提取片对高脂饮食诱导肥胖大鼠脂代谢、骨骼肌脂联素受体1(AdipoR1)和过氧化物酶体增殖物激活受体a(PPARa)基因表达的影响,以探讨大黄的减肥作用机理。方法:给饮食诱导肥胖大鼠灌服低、中、高剂量(200,400,800mg.kg-1.d-1)大黄提取片,测定血脂、脂联素,应用Real-time PCR检测大鼠骨骼肌中AdipoR1和PPARa mRNA表达水平。结果:大黄提取片呈剂量依赖性的增强降脂作用,高剂量大黄提取片能显著降低肥胖大鼠血总胆固醇(CHO)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、游离脂肪酸(FFA),显著增加骨骼肌组织PPARa mRNA表达水平,但3种剂量大黄均不能明显改变脂联素和骨骼肌AdipoR1mRNA水平。结论:大黄提取片可通过增加肥胖大鼠骨骼肌PPARa表达、降低血脂水平而促进机体减肥。 Objective: To investigate the effects of Refined Rhubarb on hpid metabohsm and genes expression of adiponectin receptorl ( AdipoR1 ) and peroxisome proliferator-activated receptor a (PPARa) in skeletal muscle of high fat diet induced obese rat, and further explore anti-obesity mechanism of Rhubarb. Methods: High fat diet induced obese rats were administrated with different dosage (200, 400, 800mg·kg^-1·d^-1) for two months. Lipid metabolic parameters and serum adiponectin were measured; the expression levels of AdipoR1 and PPARa mRNA were determined by Real-time PCR. Results: Refined Rhubarb dose-dependently lowered lipid level. High dose of Refined Rhubarb treatment decreased serum total cholesterol (CHO), triglyceride (TG), low density lipoprotein cholesterol (LDL-C), free fat acid (FFA) and increased expression of PPARa mRNA in skeletal muscle significantly, while serum adiponectin and AdipoR1 mRNA in skeletal muscle were not altered. Conclusion: Refined Rhubarb has significant anti-obesity effect via up-regulating PPARa expression and reduced lipid parameters.
