hIL-18基因转导人肺癌细胞及其肿瘤原性的研究

摘要 目的建立人白细胞介素-18(h IL-18)基因转导的肺癌细胞株,并研究其肿瘤原性。方法将携带h IL-18的质粒pcDNA 3.1-h IL-18转导入人肺癌细胞系PG细胞中,G-418进行筛选,采用RT-PCR和EL ISA法检测转导的PG细胞h IL-18的表达水平;将转导的PG细胞接种于裸鼠背部皮下,观察成瘤情况。结果h IL-18基因成功转入PG细胞中并顺利表达;h IL-18基因在mRNA水平有表达;106个转导细胞在24 h内分泌h IL-18的水平为(186.3±15.7)pg;空载体转染细胞未检测到h IL-18;裸鼠接种h IL-18基因转导的肿瘤细胞后无肿瘤长出。结论h IL-18基因转导的PG细胞肿瘤原性下降,可作为治疗肺癌的肿瘤疫苗。 [Objective] To study the effect of interleukin-18 gene transfection on the tumorigenesis of lung cancer cell line PG. [Methods] Human lung cancer cell line PG were transfected with lipofectamine pcDNA3.1- hIL-18 and selected by G418, the biologieal expression of hIL-18 was tested by RT-PCR and ELISA method; nude mice were injected subcutaneously with PG cell with or without the hIL-18 gene. [Results] The hIL-18 cDNA was sueeessfully integrated into PG cells, it was (186. 3±15. 7) pg hIL-18 secreted by one million transduced cells in 24 hours . No hIL-18 was found in cells transfected with empty vector. The nude mice injeeted with IL-18 gene engineered PG cells had no tumor growth. [-Conelusion] Tumorigenicity decreases in the IL-18 gene engineered PG cell line. The PG cells transduced with IL-18 gene might be used as lung cancer vaccine.
