摘要
目的探讨调节性T细胞及TGF-β在系统性红斑狼疮(SLE)发病中的作用机制。方法采用流式细胞仪检测54例SLE患者(SLE组)及20例健康人(对照组)外周血CD4+CD2+5T细胞和CD4+CD25h ighT细胞比率,EL ISA法定量检测血清TGF-β表达水平。结果①SLE组初发SLE者CD4+CD2+5T细胞和CD4+CD25h ighT细胞比率显著低于对照组,抗ds-DNA(+)者CD4+CD25h ighT细胞比率低于对照组,抗ds-DNA(-)者CD4+CD2+5T细胞和CD4+CD25h ighT细胞比率与对照组比较无明显差异;②SLE组TGF-β水平较对照组显著降低,其中抗ds-DNA(+)者较抗ds-DNA(-)者降低;③CD4+CD2+5T细胞与抗C1q抗体呈负相关,与TGF-β无明显相关性。TGF-β的表达量与血红蛋白量、血小板量和红细胞量呈正相关。结论调节性T细胞明显降低可能参与了SLE的发病,其免疫抑制功能的发挥可能有赖于TGFβ-。
[-Objective] To investigate the proportions of regulatory T cells, the level of TGF-β in recentonset systemic lupus erythematosus (SLE), and there roles in the pathogenesis of SLE. [Methods] Flow cytometric analysis was employed for the proportions of CD4+CD25+ regulatory T cells and CD4+CD25high T cells in the peripheral blood of SLE patients (n=54) and healthy volunteers (n=20), ELISA was employed for the level of TGF-β in blood serum. [Results] ①Recent-onset SLE group: The proportions of CD4+CD25+ cells and CD4+CD25highT cells were significantly lower than those in healthy controls;The proportions of CD4+CD25+highT cells in anti-dsDNA (+) group was lower than that in healthy controls there was no significant difference in the proportions of CD4+CD25+T cells and CD4++CD25highT cells in anti-dsDNA (-) group compared with healthy controls.②The level of TGF-β in SLE group was lower than that in healthy controls and the level of TGF-β in anti-dsDNA (+) group was lower than that in anti-dsDNA (-) group. ③CD4+CD25+ T cells was negtively correlated with anti-Clq. No significant correlation was found between levels of CD4+CD25+T regulatory cells and TGF-β. TGF-β was positively correlated with Hb, PLT numbers and RBC numbers. [Conclusion] The regulatory T cells in peripheral blood from patients with SLE is significantly decreased in comparison with healthy subjects, and they may play a role in the pathogenesis of SLE. TGF-β may be responsible for immune suppression of regulatory T cells.
