摘要 ADF蛋白是一种在真核生物中广泛存在的低分子量的肌动蛋白结合蛋白,在细胞分裂、细胞运动、花粉管伸长等重要的生命活动中发挥着重要的作用.我们以陆地棉(Gossypium hirsurum)叶片DNA为模板,通过聚合酶链式反应(PCR)扩增分离获得棉花GhADF7基因.该基因长度为902bp,包含420bp阅读框;编码区含有3个外显子和2个内含子;其编码的ADF蛋白含有139个氨基酸残基.GhADF7与其它生物的ADF蛋白具有较高的同源性.其中GhADF7与NtADF1,NtADF2的亲缘关系较近,与LIADF、ZmADF的亲缘关系次之,它们的氨基酸序列同源性在75%~79%之间.上述系统发育进化分析表明,植物ADF基因具有高度保守性. ADF (actin depolymerizing factor), highly conserved in all eukaryotic cells, is a low molecular mass actin-binding protein. It plays a key role in the process of cell division, cell movement, pollen tube elongation and so on. Using the DNA from cotton (Gossypium hirsutum) leaves as template, GhADF7 gene were isolated by PCR. The GhADF7 gene is 902bp in length, containing two introns. The gene deduced an ADF protein with 139 amino acid residues. Analysis of evolution relationship revealed that GhADF7 is more closely related to LIADF and ZmADF than to NtADF1 and NtADF2. Their homology is between 75%- 79%. The analysis of phylogenetic relationship showed that plant ADF genes are highly conserved on structure during evolution.
