摘要
目的探讨肌-内皮连接偶联系统在移植静脉重塑早期对血管平滑肌细胞(VSMC)表型转化态的影响。方法20只大鼠随机分为实验组(基因转染组)与对照组(非基因转染组),于术后14d取出移植静脉,采用反转录多聚酶链反应(RT-PCR)及电镜等检测并观察缺氧诱导因子-1α(HIF-α)表达水平、肌-内皮连接结构、血管内皮细胞(VEC)及血管平滑肌细胞(VSMC)表型。结果实验组较对照组HIF-Iα过表达,VEC增生显著,VSMC以收缩表型居多,肌-内皮连接结构重建显著(P均<0.01)。结论肌-内皮连接偶联系统是稳定移植静脉重塑早期VSMC转化态重要的组织结构和评估血管结构与功能良好重建的重要标志物之一。
Objective : To explore the effeots of couple system of myo-endothelial junotion on phecotyplc chanses of vascular smooth muscle cells in the process of vascular remodeling esrly. Methods :20 Wistar rats weishing 250-3008 were randomly divided into two Stoups: control group and experimental Stoup, 10 rats in each Stoup. A rat experimental model of autosenous vein graft was established by transplanting the risht external jusular into the right common carotid artery in 80 rats. The transplanted vein in the experimental group was immersed using recombinant edeno-associated virus(rAAV)HIF-1α solution for 15 minutes before anastomosis,while the controle ones was immersed only in recombinant adeno-assoiated virus solution without HIF- 1α gene for 15 minutes. The vein grafts samples were taken at the 14^th day after operation. Reverse transcription polymerase chain reaction (RT-PCR) and Western blotting technique were used to detect the expression of HIF-1α mRNA and protein. Results :There was remarkable Increase of expression level of HIF-1α mRNA and VEGF protein than that in control Stoup. The vascuterculer endothella cell and myo-endothelium junction in vein grafts were remodeled at the 14^th day after operation In the experimental Stoup. constrictive phenotype of vascular smooth muscle oell were found in 68.7% of the experimental group. Conclusion : The couple system of myo-endothella junction on the stabilization of phenotype chanse of vascular smooth muscle cell is one of the valuable vascular structures,and plays a crucial role in the process of vascular remodeling early,and a marker to evaluate a reconstruction about the function and tissues in venous autografts In rat.
