摘要
目的研究小檗碱对高转移性人巨细胞肺癌PG细胞黏附特性的影响,以探讨其抑制肿瘤侵袭转移的可能机制。方法小檗碱(5、10μg/ml)处理PG细胞24小时,采用MTT法观察PG细胞与PG细胞的黏附(同质黏附),PG细胞与细胞外基质(FN和Martrigel)的黏附;用虎红染色法观察PG细胞与人脐静脉内皮细胞(HUVEC)的黏附。通过SABC免疫细胞化学法及图像分析软件,半定量PG细胞E-cadherin、CD44的表达;RT-PCR法检测PG细胞E-cadherinmRNA和CD44mRNA的表达。结果小檗碱(10μg/ml)可促进PG细胞的同质黏附(P<0.05);小檗碱(5、10μg/ml)可抑制PG细胞与FN和Mar-trigel的黏附(P<0.01),并可抑制PG细胞与HUVEC的黏附(P<0.05,P<0.01)。经小檗碱(10μg/ml)作用的PG细胞,E-cadherinm及其mRNA表达明显增高(P<0.01),而CD44及其mRNA的表达明显减少(P<0.05)。结论小檗碱通过促进PG细胞E-cadherin的表达而促进PG细胞间的同质黏附,抑制PG细胞CD44的表达而抑制其与细胞外基质及血管内皮细胞的黏附,这可能是小檗碱抗肿瘤转移的机制之一。
Objective:To study the effect of berberine on adhesion characteristic of PG cell line and to investigate POssible mechanism of inhibiting tumor metastasis of berberine. Methods : MTT assay was used to detect PG-PG homotypie adhesion and PG-matrix (FN, matrigel) adhesion,and PG-HUVEC adhesion was determined by Rose Bengal stain. Auantitative immunoeytoehemieal method and RT-PCR were applied to detect the expression of E-eadherin and CD44 protein and mRNA respectively. Results:After 24 h treatment, berberine enhanced PG-PG homogeneous adhesion at 10μg/ml (P 〈 0. 05 ) and reduced PG-matrix(FN,matrigel) adhesion and PG-HUVEC adhesion at 5,10 μg/ml(P 〈0. 05 ,P 〈0. 01 ). The expression of E-cadherine protein and mRNA was increased by berberine, while CD44 was decreased. Conclusion : Berberine enhances PG-PG homogeneous adhesion through increasing the expression of E- cadherine protein and mRNA and weaks PG-matrix and PG-HUVEC adhesion through decreasing the expression of CD44 protein and mRNA, which may be one of mechanisms of inhibiting tumor metastasis of berberine.
