黄芩苷抗肺炎衣原体致动脉内皮细胞损伤研究 被引量:9

The Effect of Baicalin on Protecting Endothelial Cells

摘要 目的:探讨肺炎衣原体(CPn)感染对主动脉内皮细胞的损伤作用及黄芩苷的干预作用。方法:小鼠随机分为黄芩苷高剂量组、黄芩苷低剂量组、阿奇霉素组、模型组、正常对照组。喂饲高胆固醇饲料1周后,经一侧鼻腔吸入含CPn的培养液,每周1次,共3次。最后1次接种5天后开始给药至实验结束,第1次接种后第18周处死全部动物。取主动脉检测血管反应性、电镜检测内皮细胞(EC)损伤和动脉粥样硬化斑块细胞凋亡。结果:模型组主动脉对内皮依赖舒张反应降低,细胞凋亡指数、内皮细胞损伤程度升高,3个给药组以上3个指标均趋向正常。结论:黄芩苷和阿奇霉素具有保护血管内皮细胞从而阻断AS发病过程的作用。 Objective: To study the influence of CPn infection on aortic endothelial cells and the intervention effect of baicalin. Methods :Eight- week- old SPF male C57BL/6J mice were divided randomly into 6 groups : high - dosage - baicalin group ( group Ⅰ ), low - dosage - baicalin group ( group Ⅱ ), azithromycin group (group Ⅲ ) , model group ( group Ⅳ ) , and normal group ( group Ⅴ ). A week after being fed cholesterol - supplemented diet, group Ⅰ , group Ⅱ , group Ⅲ, group Ⅳ and group Ⅴ were inoculated with CPn at (0.5 - 1 ) × 10^7 IFU per inoculation. Each mouse of these groups was inoculated once weekly for 3 consecutive weeks. Administration started 5 days after the last inoculation. Rusults :In model group, endothelium dependent relaxation caused by acetylcholine was reduced, and cell apoptosis index and the injure degree of endothelial cells increased. Whereas, endothelium dependent relaxation, cell apoptosis index and the injure degree of endothelial cells trended to normal degree in the three treatment groups. Conclusion :Both baicalin and azithromycin could protect the vascular endothelial cells.
