花边莲提取液诱导人肺癌SPC-A-1细胞凋亡及其分子机制的研究 被引量:5

摘要 目的:探讨花边莲提取液对人肺癌SPC-A-1细胞凋亡及其可能分子机制。方法:采用MTT法检测细胞增殖抑制率,TUNEL法检测凋亡率,二步法免疫组化检测野生型P53、Survivin、NF-κB P65和Bc l-2蛋白表达变化。结果:花边莲提取液能够抑制SPC-A-1细胞增殖,且呈一定剂量—时间依赖性;随着药物浓度上升细胞凋亡率明显增高(P<0.01);P53蛋白表达显著上升(P<0.01),Survivin、NF-κB和Bc l-2蛋白表达明显下降(P<0.01)。结论:花边莲取液能诱导SPC-A-1细胞凋亡,其分子机制可能与上调野生型P53蛋白表达和下调Survivin、NF-κB P65和Bc l-2蛋白表达有关。 Objective : To investigate the apoptosis - inducing of HuaBianLian(HBL) extract and its possible molecular mechanism on human lung cancer SPC - A - 1 cells. Methods: The proliferation inhibitory rate was evaluated by MTr assay. Induction of cell apoptotic rate was determined by TUNEL method. The expression of wild type P^53, Survivin, NF - κBP^65 and Bcl -2 proteins were detected by two - step immunhistochemical staining. Results: HBL extract could inhibit the proliferation of SPC - A - 1 cells and inhibitory effect was dose - and - time - dependent. The rate of apoptosis increased significantly with the increasing concentrations of the drug,Tile expression of p^53 protein was increased significantly( P 〈 0.01 ), while Survivin, NF - κBP^53 and Bcl - 2 proteins were decreased markedly( P 〈 0.01 ). Conclusion : HBL can induce the apoptosis of human lung cancer SPC - A - 1 cells, and its molecular mechanism may be associated with the up - regulating expression of wild type P^53 protein and down - regulating expression of Survivin, NF - κBP^65 and Bcl - 2 proteins.
