利用荧光偏振研究二价金属离子对DNA与组蛋白结合的影响 被引量:3

摘要 利用荧光偏振实验我们研究了二价金属离子(Mn^(2+),Mg^(2+)和Ca^(2+))对DNA与组蛋白结合的影响.测量了二价金属离子(Mn^(2+),Mg^(2+)和Ca^(2+))存在时,DNA和DNA-组蛋白的荧光偏振度.结果表明,组蛋白明显降低DNA分子的荧光偏振度;二价金属离子显著增加DNA分子的荧光偏振度.与DNA和组蛋白单独培育时相比较,当二价金属离子、组蛋白、DNA共同培育时,DNA可与更多的组蛋白结合.Mn^(2+)比其他二价离子(Mg^(2+)和Ca^(2+))更容易使DNA-组蛋白复合体发生凝聚现象.
