摘要 胆固醇调节元件结合蛋白(SREBPs)是膜结合的转录因子,参与脂质自稳态的许多方面(例如胆固醇、脂肪酸、甘油三酯和磷酯的合成/摄取)。SREBPs在内质网上合成,需要被转运到高尔基体并经过一系列蛋白水解后才能到核内发挥作用。此过程是由两个蛋白介导的,SCAP和Insig。本综述旨在对SCAP和Insig在SREBP加工成熟及在脂质稳态方面的作用进行概述。 Sterol regulatory element binding proteins (SREBPs) are membrane-bound transcription factors that activate genes encoding enzymes of cholesterol and fatty acid biosynthesis. SREBPs are synthesized as precursor proteins in the endoplasmie reticulum (ER), where they form a complex with another protein, SREBP cleavage activating protein (SCAP). SCAP escorts SREBP to activation in the Golgi apparatus. There, SREBP undergoes two proteolytic cleavage steps to release the mature, biologically active transcription factor, nuclear SREBP ( nSREBP). The process is induced by two proteins, SCAP and Insig. This review is to summarize the role of SCAP and Insig in the procession and maturation of SREBP and the lipid metabolism.
