40种香豆素类化合物对人表皮癌细胞系A432细胞株和人乳腺癌细胞系BCAP细胞株增殖抑制活性的筛选 被引量:23

摘要 目的从中药中筛选有抗肿瘤活性的化合物、类药或先导化合物。方法应用人表皮癌细胞系A432细胞株和人乳腺癌细胞系BCAP细胞株筛选40种香豆素类化合物对其增殖的抑制活性。结果花椒毒素、前胡素、虎耳草素、牛防风素和甘草香豆素对人表皮癌细胞系A432细胞株增殖具有弱的抑制作用,且呈浓度-效应关系;其IC50值分别为8.40×10-4、5.00×10-5、7.30×10-5、1.00×10-5、5.00×10-5mol.L-1。蛇床素、欧前胡素、香柑内酯、羌活烯醇、紫苜蓿酚和考迈斯托醇对人乳腺癌细胞系BCAP细胞株增殖具有弱的抑制作用,且呈浓度-效应关系;其IC50值分别为6.70×10-5、2.11×10-4、2.77×10-5、6.62×10-5、4.16×10-5、2.26×10-4mol.L-1。结论某些香豆素类化合物具有潜在的抗肿瘤活性,可能发展成为类药或先导化合物。 Objective: To screen Medicines. Methods: Forty coumarins antitumor active, drugs - like or leading compounds from Traditional Chinese compounds isolated from the Traditional Chinese medicine were studied for their antitumor activity in vitro by determining the inhibitory percentage against growth of human epidermal carcinoma cell line A432 and human mammary cancer cell line BCAP, respectively. Results: It showed that xanthotoxin, decursin, pimpinellin, sphondin and glycycoumarin inhibited growth of human epidermal carcinoma cell line A432 with IC50 values of 8.40×10^-4、5.00×10^-5,7.30×10^-5、1.00×10^-5、5.00×10^-5mol·L^-1, respectively, and their activity showed concentration - effect relationship. Cnidilin, imperatorin, bergapten, notoptol, dicoumarol and coumestrol inhibited growth of human mammary cancer cell line BCAP with IC50 values of 6.70×10^-5、2.11×10^-4、2.77×10^-5、6.62×10^-5、4.16×10^-5、2.26×10^-4mol·L^-1, and their activity showed dose - effect relationship. Conclusion: Some coumarins compounds are potential anti - tumor agent and may be developed for drugs -like or leading compounds.
