摘要
目的探讨重组人骨形态发生蛋白-7(rhBMP-7)对大鼠心肌缺血再灌注损伤的保护作用。方法建立W istar大鼠心肌缺血再灌注(I-R)模型,随机分为对照组(C组)、I-R(I组)和rhBMP-7处理组(B组),B组于血流阻断前10 m in股静脉给予rhBMP-7 250μg/kg,C组和I组给予等量生理盐水。于血流再灌注后24 h取血检测乳酸脱氢酶(LDH)和磷酸肌酸激酶(CPK)含量,心肌匀浆检测丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性,TTC法测定心肌梗死面积,电镜下观察心肌组织学变化。结果rhBMP-7处理组大鼠心肌I-R24 h后,其心肌酶含量、氧自由基MDA、心肌梗死面积,均明显低于I组;SOD活性增加(P<0.01)。心肌细胞的坏死程度也明显轻于I组。结论rhBMP-7缩小梗死面积,对心肌I-R损伤具有保护作用。可能与其抑制脂质过氧化,保护抗氧化酶活力有关。
Objective To investigate the protective effect of recombinant human bone morpbegenetic protein-7 (rhBMP-7) on myocardial ischemia-reperfusion (IB) injury of rats. Methods Wistar rats of myocardial IR model were randomly divided into IB group (250 μg,/kg saline), rhBMP-7 group (250 p,g/kg rhBMP-7 via femoral vein 10 rain before blood flow interruption) and control group (250 μg/kg saline). The lactate dehydrogenase (LDH) and creatine phosphokinase (CPK) contents in blood, malondialdehyde (MDA) contents and superoxide dismutase (SOD) activity in myocardial homogenate were detected 24 h after IR. Infarct sizes were assessed by TIC method. Histological findings were observed by electron microscope. Results In rhBMP-7 group, LDH, CPK, MDA and infarct size were lower than those in IR group; SOD were higher than that in IR group (P 〈 0. 01 ) ; myocardial cell necrosis was slighter than that in IR group. Conclusions rhBMP-7 has a protective effect on myocardial IR injury through lessening infarct size, which might be related to inhibiting lipid peroxidation and protecting antioxidsse activity.
