摘要
介绍一种简便快速提取细菌质粒的改进方法,不需要按常规对细菌培养物离心和用溶液Ⅰ、Ⅱ和Ⅲ处理,而是直接用酚-氯仿(1∶1)处理,离心分层取上清,异丙醇沉淀,所得质粒产量高,纯度足以用于酶切。
A simple and rapid method to isolate plasmid from bacteria is introduced. Bacteria culture was treated by Phenol -CHCl3 （1 ： 1 ）, upper aqueous phase was withdrawnand plasmid DNA was precipitated by isopropanol. The routine steps such as colleetinghacteria by centrifugation and treating with Solution Ⅰ , Ⅱ and Ⅲ were thus avoided, The plasmid obtained was pure enough for enzyme digestion.
