滨蒿内酯对豚鼠哮喘模型的炎性介质抑制作用 被引量:14

摘要 目的:研究滨蒿内酯(Sco)对豚鼠哮喘的抗炎作用及机制。方法:制备迟发性哮喘豚鼠模型,免疫组化法检测豚鼠血浆和支气管肺泡灌洗液(BALF)中白介素5(IL-5)含量;电镜法对豚鼠肺组织细胞溶酶体磷脂酶A2(PLA2)进行染色,并通过图像分析定量检测其含量。结果:哮喘组IL-5及PLA2的含量与正常对照组比较明显增高(P<0.05);提前雾化给予42 mg/L,340 mg/L的Sco组与哮喘模型组比较明显降低IL-5及PLA2含量(P<0.05)。结论:Sco可能通过抑制IL-5及PLA2发挥其对支气管哮喘的作用。 Objective: To study the anti-inflammatory effect of scoparone (Sco) on asthma and its mechanism in guinea pigs. Methods: Guinea pig model of late asthmatic reaction (LAR) was established. The content of interleukin-5 (IL-5) in serum and bronchial alveolar lavage fluid (BALF) was detected by using immunohistechemical method. The content of phospholipase A2 (PLA2 ) in the lysosomes of pulmonary tissues was detected by image analysis after staining. Results: The contents of IL-5 in serum and BALF and PLA2in the lysosomes of pulmonary tissues in the asthma group were significantly higher than those in the control group ( P 〈 0.05). The contents of IL-5 and PLA2 iu tbe treatment group (Sco of 42 and 340 mg/L) were significantly lower than tbose in the asthma group ( P 〈0.05). Conclusion: Sco can inhibit inflammation by lowering the contents of IL-5 in serum and BALF and PLA2 in pulmonary tissues in asthmatic guinea pig model.
