摘要
目的:研究肿瘤坏死因子相关凋亡诱导配体(TRAIL)对骨肉瘤OS-732细胞的诱导凋亡作用,探寻骨肉瘤临床化疗的新方案。方法:将TRAIL应用于体外培养的骨肉瘤OS-732细胞,采用MTT法检测细胞毒性作用,倒置相差显微镜及荧光染色方法观察肿瘤细胞形态改变,扫描及透射电镜在亚细胞形态上证实凋亡细胞。结果:不同浓度TRAIL作用下,细胞抑制率间差别十分显著(P<0.01)。超微结构观察显示骨肉瘤细胞凋亡。结论:TRAIL诱导骨肉瘤细胞凋亡,且具有浓度依赖性。
Objective: To study the role of TNF-related apoptosis-inducing ligand (TRAIL) in the apoptosis of osteosarcoma cell line (OS-732) and explore the new chemotherapeutic strategy. Methods: TRAIL was administered to OS-732 cells cultured in vitro, and the inhibition rate of OS-732 cells was measured by using MTT assay. The cellular ultrastructure was observed under inverted phasecontrast microscope, fluorescence microscope, and scanning and transmission electron microscope. Results: There was significant difference in the inhibition rate among OS-732 cells under different concentrations of TRAIL 24 hours after administration of TRAIL ( P 〈 0.01 ). The changes of cellular ultrastructure showed TRAIL induced the apoptosis of OS-732 cells. Conclusion: TRAIL could induce the apoptosis of osteosarcoma ceils in a dose-dependent manner.
