摘要
目的:探讨氧化苦参碱的抗I型变态反应的作用机制。方法:用MTT比色法检测不同浓度的氧化苦参碱对RBL-2H3细胞活化的影响。根据MTT比色法分别选择氧化苦参碱5个不同的实验浓度,并以色甘酸钠作为阳性对照。以C48/80刺激RBL-2H3细胞活化脱颗粒,测不同浓度药物对RBL-2H3细胞分泌组胺及TNF-α变化。结果:氧化苦参碱对RBL-2H3细胞的增殖有抑制作用。氧化苦参碱能抑制RBL-2H3细胞释放组胺及TNF-α,抑制作用不弱于色甘酸钠组。氧化苦参碱浓度的对数与RBL-2H3细胞释放组胺和TNF-α抑制率呈高度线性关系。结论:氧化苦参碱能抑制RBL-2H3细胞分泌组胺和TNF-α,该抑制作用呈剂量依赖性。其治疗I型变态反应性皮肤病可能和它抑制肥大细胞活化脱颗粒有关。
Objective: To determine the direct effect of oxymatrine on the degranulation function of RBL- 2H3 mast cells. Methods: ( 1 ) The proliferation of RBL - 2H3 mast cells induced by oxymatrine at different concentrations was determined with MTT colorimetric assy. (2) Pretreated RBL - 2H3 cells with different dilutions of oxymatrine were activated by compound C48/80. The tumor necrosis factor- alpha (TNF- α) from the supernatants, the spontaneously released histamine and the total histamine were measured by ELISA. Results: (1) Oxymatrine suppressed the proliferation of RBL- 2H3 mast cells. (2) The suppressed percentage of the histamine and TNF- α released from RBL- 2H3 mast cells in oxymatrine group was no comparable to sodium cromoglicate control group and in a concentration dependent manner. The suppressed percentage of the histamine and TNF - α was hnearly correlated with the concentration of oxymatrine. Conclusion: Oxymatrine - suppressed degranulation of RBL - 2H3 cells may be one of the mechanisms of type - Ⅰ anti - allergic reaction in the treatment of skin diseases.
