摘要
目的探讨球后注射脂质体包裹的激活蛋白-1诱骗寡核苷酸(activator protein-1decoy oligodeoxynucleotides,AP-1decoy ODN)对豚鼠近视眼屈光状态的影响。方法人工合成AP-1decoy ODN及错配的AP-1decoy ODN的核苷酸序列。半透明眼罩遮盖单眼诱导豚鼠近视眼的形成。遮盖眼球后注射脂质体包裹的AP-1decoy ODN或错配的AP-1de-coy ODN,检影验光测眼球屈光度,用A超(20Hz)测眼轴长度。结果豚鼠遮盖眼眼轴延长,形成近视。球后注射AP-1decoy ODN+脂质体LipofectAMNETM的遮盖眼近视程度减轻,而球后注射错配AP-1decoy ODN+脂质体LipofectAMNETM对遮盖眼的近视程度无影响。结论球后注射脂质体包裹的AP-1decoy ODN能有效抑制豚鼠近视眼的形成。
Objective To investigate the effect of retrobulbar injection of liposome-mediated activator protein-1 decoy oligodeoxynucleotides (AP-1 decoy ODN) on the myopic eye of the guinea pig. Methods AP-1 decoy ODN and scrambled AP-1 decoy ODN were synthesized. Form-deprivation myopia was induced in the guinea pig by a translucent diffuser worn over one eye. Liposome-mediated AP-1 decoy ODN or scrambled AP-1 decoy ODN was injected retrobulbarly into the eyes wearing a diffuser. Ocular refraction and axial length were assessed by retinoscopy and A-scan ultrasonography (20 Hz). Results The guinea pig eye wearing the diffuser elongated axially and became myopic. Retrobulbarly injected AP-1 decoy ODN containing liposomes (LipofectAMNE^TM) could ameliorate the myopic refractive change induced by form deprivation. However, the. myopic refractive state did not change in the deprived eyes that had been injected with scrambled AP-1 decoy ODN containing liposomes. Conclusion Retrobulbarty injected AP-1 decoy ODN could significantly inhibit the development of myopia in the guinea pig eye.
