胃癌及其转移淋巴结组织中胰岛素样生长因子-I的表达及其临床意义

摘要 目的探讨IGF-I在胃癌发生发展中的作用及其临床意义。方法应用荧光定量RT-PCR法,检测40例胃癌及其转移淋巴结组织中的IGF-I表达情况,以40例消化道溃疡胃组织标本作对照,将其表达结果和患者临床病理指标进行综合分析。结果胃癌中IGF-I的表达[(9.24±3.16)×104拷贝/μgRNA]明显高于正常胃黏膜组织[(2.09±1.05)×103拷贝/μgRNA](P<0.05);TNM分期中Ⅰ+Ⅱ期IGF-I表达[(7.35±1.24)×104拷贝/μgRNA]低于Ⅲ+Ⅳ期[(1.05±3.44)×105拷贝/μgRNA](P<0.05);癌转移淋巴结组织IGF-I表达[(1.03±0.36)×105拷贝/μgRNA]明显高于无癌转移淋巴结[(7.62±1.17)×104拷贝/μgRNA](P<0.05)。结论IGF-I在胃癌组织中的高表达可能与胃癌的发生、发展有关;癌转移淋巴结组织中IGF-I表达量明显增高,提示IGF-I有可能参与了胃癌的侵袭和转移。 Objective To investigate the expression of IGF-Ⅰ in gastric carcinomas and metastatic lymph nodes,so as to explore the mechanism of gastric carcinogenesis and clinical significance. Methods Forty patients with primary gastric cancer,40 with gastroduodenal ulcers were included to determine the IGF-Ⅰ expression by using FQ-PGR. The levels of the expression were correlated with clinicopathological factors. Results The mRNA levels of IGF-Ⅰ were increased in gastric cancer tissues[ (9.24 ± 3.16) × 10^4 copies/μg RNA] compared with normal tissues (2.09± 1.05 ) × 10^3 copies/μg RNA from healthy controls ( P 〈 0.05) ; There was a significant difference of the levels of IGF-Ⅰ between the group of TNM Ⅰ+Ⅱ [(7.35 ± 1.24) × 10^4 copies/μg RNA] and Ⅲ + Ⅳ [ (1.05 ± 3.44) × 10^5 copies/μg RNA] (P 〈 0.05) ;ThemRNA levels of IGF-Ⅰ in metastatic lymph nodes [ (1.03 ± 0.36) × 10^5 copies/μg RNA] were higher than in without metastatic lymph nodes[(7.62 ± 1.17) × 10^4 copies/μg RNA]. Conelnsion The expression levels of IGF-Ⅰ in gastric cancer were higher than that in normal tissue(P 〈 0.05),which suggested that the higher IGF-Ⅰ expression was correlated with the genesis of gastric cancer; The mRNA levels of IGF-Ⅰ were higher in the group of TNM Ⅲ +Ⅳ and metastatic lymph nodes,which suggested that IGF-Ⅰ was associated with gastric cancer invasion and metastasis .
