外源性L-精氨酸对小鼠日本血吸虫病肝脏内I型和Ⅲ型胶原的影响

摘要 目的:动态观察外源性L-精氨酸对小鼠日本血吸虫病肝脏内I型和Ⅲ型胶原的影响,探讨其治疗血吸虫病肝纤维化的可能性。方法:随机将54只健康小鼠分为正常组、感染组和L-精氨酸组。感染组和L-精氨酸组小鼠经腹部皮肤感染日本血吸虫尾蚴。L-精氨酸组于感染后第6周将1 g/L的L-精氨酸水溶液给小鼠饮用。于感染后的第9、11、13、15周每组随机挑选6只处死取肝脏做病理观察;并对随机选取的30个视野下I型和Ⅲ型胶原面积及两者之比值作定量分析。结果:感染组与L-精氨酸组小鼠肝脏内I型和Ⅲ型胶原面积在感染第9周后逐渐减小,I型和Ⅲ型胶原面积比值变化较大;L-精氨酸组I型和Ⅲ型胶原面积比感染组减小明显(P<0.05)。结论:L-精氨酸具有减少日本血吸虫病肝脏组织内I型胶原和Ⅲ型胶原的作用。 Aim: Through observing the effects of L- Arginine on Collagen Ⅰ and Collagen Ⅲ in the hepatic tissues of mice with Schistosoma japonicum, to explore its action of curing liver fibrosis. Methods: 54 mice were randomly divided into normal group (6 mice), infective group (24 mice) and L- Arginine treatment group (24 mice). Each mouse both in infective group and L- Arginine treatment group was infected with cercaria of Schistosoma japonicum through its skin of abdomen. 1 g/L L - Arginine solution was freely drunk by the mice of L - Arginie treatment group in the 6^th week after infected. 6 mice from each group were killed at 9^th, 11^th, 13^th and 15^th weeks respectively after infected and their livers were collected to undergo staining and pathologic observation by micropolarimete. The areas of Collagen Ⅰ and Collagen Ⅲ in hepatic fibroses were measured using computer image analysis software. Results: the areas of Collagen Ⅰ and Collagen Ⅲ in the hepatic tissues of mice both infective group and L - Arginine treatment group reduce generally, and the change of the ratio of Collagen Ⅰ and Collagen Ⅲ is large, and the areas of Collagen Ⅰ and Collagen Ⅲ in L- Arginine treatment group were reduced more obviously than in infective group ( P 〈 0. 05). Conclusion: L - Arginine has effects on promoting decrease of Collagen Ⅰ and Collagen Ⅲ in the hepatic tissues of mice with Schistosoma japonicum.
