摘要
目的探讨布地奈德对哮喘豚鼠气道重塑和支气管肺泡灌洗液(BALF)中Ⅲ型前胶原(PCⅢ)、透明质酸(HA)及层粘蛋白(LN)的影响,为糖皮质激素在哮喘中的应用提供理论依据。方法健康雄性豚鼠24只,按体质量随机分为3组,正常对照组、哮喘组和哮喘模型布地奈德雾化吸入治疗组(布地奈德组),每组8只。利用计算机图像分析仪测定支气管管腔的内周长(Pi)、支气管管壁厚度(WAt/Pi)、支气管平滑肌厚度(WAm/Pi)、支气管平滑肌细胞计数(N/Pi);放射免疫法检测BALF中细胞外基质(ECM)成分PCⅢ、HA、LN的改变。结果与正常对照组比,哮喘组WAt、WAm及N明显升高(P<0.05),BALF中PCⅢ、LN及HA含量均明显增加(P<0.05);与哮喘组比,布地奈德组气道壁结构明显改善,WAt、WAm及N明显下降(P<0.05),BALF中PCⅢ、LN及HA明显减少(P<0.05)。结论布地奈德可延缓哮喘豚鼠气道重塑的发生,这可能与其抑制ECM成分PCⅢ、LN及HA在气道中的沉积有关。
Objective To explore the effects of Budesonide on airway remodelling and contents of PCⅢ ,LN and HA in bronchoalveolar lavage fluid (BALF) in asthmatic guinea-pigs. Methods Twentyfour healthy male guinea-pigs were divided into three groups according to weight: the normal control group(n=8), asthmatic group(n=8) and budesonide(BUD) inhalation group(n=8). The airway wall thickness(WAt/Pi), bronchial smooth muscle thrickness(WAm/Pi), bronchial smooth nucleus counts (N/Pi) were measured by using computer-assisted image analysis system. The contents of PC Ⅲ, LN and HA in BALF were detected by radioimmunity. Results Compared with the normal control group, WAr,WArn and N in bronchus and the contents of PCⅢ ,LN and HA in BALF in asthmatic group were all significantly increased. Compared with the asthmatic group, the structure of the airway wall was ameliorated in budesonide inhalation group, WAt,WAm and N in bronchus and the contents of PCⅢ, LN and HA in BALF were all significantly decreased. Conclusion Budesonide can delay the course of the airway remodeling and this may be related with its roles of suppressing the PC Ⅲ, LN and HA deposition in airway.
