摘要 目的:观察缓激肽(BK)对大鼠肾系膜细胞增殖和纤维连接蛋白表达的影响。方法:采用体外培养的大鼠肾系膜细胞,应用四甲基偶氮唑盐(MTT)比色法检测细胞增殖情况;应用Westernblotting观察肾系膜细胞纤维连接蛋白的表达。结果:缓激肽促进肾系膜细胞增殖,且具有剂量依赖性。缓激肽和转化生长因子-β1皆显著增加系膜细胞纤维连接蛋白表达,并具有协同效应。结论:缓激肽可能直接促进体外培养的系膜细胞增殖和纤维连接蛋白表达,从而加重对肾小球的损伤。 Objective: To investigate the direct effect of hradykinin(BK) on proliferation and fibronectin expression in cultured rat mesangial cells and whether the effects of BK and transforming growth factor-β1 (TGF-β1) on fibronectin levels are synergistic. Methods: Mesangial cell proliferation was assessed by MTT assay, and fibronectin expression in mesangial cells was investigated by Western blotting. Results: In quiescent mesangial cell cultures (0.5% serum for 24 hr), BK stimulated cell proliferation in a dose dependent fashion. Both BK and TGF-β1 induced a significant increase in mesangial cell fibronectin levels. Furthermore, the effects of BK and TGF-β1 on fibronectin levels were synergistic. Conclusion: These results suggest that BK directly stimulates proliferation and fibronectin expression in cultured mesangial cells, thus promoting glomerular injury.
