杂交稻种子纯度的SSR分子检测及应用研究 被引量:19

摘要 利用SSR标记构建了15份恢复系和11份不育系共26份籼型杂交水稻亲本的DNA指纹图谱,筛选出了冈优725和Ⅱ优838两个杂交种亲本的共显性SSR标记。同时对四川省6家种子企业生产的两个杂交水稻品种冈优725、Ⅱ优838及其亲本进行田间和SSR标记纯度鉴定。结果表明两种鉴定方法无显著差异,均可应用于种子纯度检测,但分子标记纯度检测的结果更准确、可靠。不同来源的种子纯度存在显著差异,6家种子企业提供的亲本及杂交种的纯度普遍较低,多数未达国家标准。 Utilization of hybrid rico plays an important role in increasing crop yield. Seed quality is the most important factor one of improving yield of hybrid rice, and the core of which is seed purity. With the rapid development of crop breeding, seed purity could not be tested easily by using traditional methods such as phenotype. So it requires better method to identify more and more combinations. In the investigation reported in this article, the fingerprinting map is constructed with SSR of 26 Indica hybrid parents including 15 restorers and llsterile lines. According to the fingerprinting map, primers were selected to test purity of F1 hybrids, Gangyou725, Ⅱ-you838 and their parents from six seed companies. Meanwhile compared with the result of field identification, there' s no significant difference between the two methods, and both are applied into purity test. But the result by means of SSR molecular markers is more exact and reliable. The method can be employed in any season, which is a developing trend to identify seed purity and authenticity. In the analysis of seed purity from different resources, it was showed that significant differenco existed, In addition, seed purity from different companies almost could not reach national standard.
