摘要
目的:探讨人脑胶质瘤组织缺氧诱导因子1α(HIF-1α)表达与肿瘤血管生成和恶性程度的相关性。方法:对手术获得的不同恶性程度的人脑胶质瘤标本,应用免疫组化和RT-PCR方法,检测HIF-1α和血管内皮生长因子(VEGF)在蛋白和mRNA水平的表达程度,并进行统计学分析。结果:不同恶性程度人脑胶质瘤HIF-1α的蛋白和mRNA表达有差异,与肿瘤恶性程度呈正相关关系。VEGF表达有相同趋势,并与肿瘤HIF-1α表达呈正相关关系。结论:随人脑胶质瘤恶性程度增高。HIF-1α表达增强,从而促进肿瘤血管生成,促进肿瘤生长。
Objective: To investigate the expression of hypoxia-inducible factor 1α(HIF-1α) in human gliomas and its correlation between it and tumor angiogenesis and pathological grades. Methods: Using immunohistochemical staining and RT-PCT, HIF-1α and VEGF of tissues with different grades gliomas were measured on protein and mRNA level, Results: There was significant difference of the HIF-1α expression level in different grades of human gliomas, Positive correlation was found between the expression level and tumor grade. The VEGF expression was similar to HIF-1α, and had positive correlation with it. Conclusion: The expression of HIF-1α increases in high grade gliomas, and it promotes the tumor angiogenesis and growth.
