山楂叶总黄酮对大鼠脑缺血再灌注后基质金属蛋白酶-9表达的影响 被引量:11

摘要 目的研究山楂叶总黄酮对大鼠脑缺血再灌注损伤中基质金属蛋白酶-9(MMP-9)表达的影响。方法雄性Wistar大鼠,随机分为假手术组、缺血再灌注组及山楂叶总黄酮组(60、30、15mg/kg),线栓法建立大鼠大脑中动脉闭塞局灶性脑缺血再灌注模型。观察再灌注3d后脑梗死面积,通过免疫组织化学染色方法测定MMP-9的表达。结果山楂总黄酮组(60、30mg/kg)的脑梗死面积较缺血再灌注组明显减少,MMP-9的表达明显低于缺血再灌注组,P<0.01。结论山楂叶总黄酮能明显地减少脑梗死的面积,降低MMP-9的表达,发挥脑保护作用。 Objective To investigate the effect of hawthorn leaves flavonoid(HLF)by the MMP-9 expression of brain ischemia and reperfusion injury in rats. Methods Male Wistar rats were randomly divided into sham-operated group,ischemia-reperfusion group and HLF treatment groups (60,30,15mg/kg). Rat models of focal ischemiareperfusion of middle cerebral artery occlusion(MCAO) were established by using thread method,the MMP-9 expressions were assayed using immunohistochemistry staining method and cerebral infarction areas were calculated after 3 days brain ischemia and reperfusion injury. Result There were smaller cerebral infarction in the groups of HLF treatment groups(60,30mg/kg) than ischemia-reperfusion groups, the expressions of MMP-9 in therapeutie groups (HLF 60,30mg/kg) are lower than ischemia-reperfusion groups, P 〈 0. 01. Conclusions The treatments with HLF could diminish cerebral infarction area,depress the expression of MMP-9, play the roles of cerebral protection.
