摘要
目的探明Orexin A对麻醉大鼠皮层调节作用的间接途径及可能机制。方法将55只大鼠分为4组:①对照组;②Orexin A组;③Pyrilamine组;④Pyrilamine+Orexin A组。用玻璃微电极记录4组大鼠前额叶皮层脑区神经元细胞外电活动。结果①Orexin A溶液可明显增加额叶皮层神经元的发放频率(n=12),在6min内达到高峰,与基础值比较,给药结束后前9min各时间段明显增加(P<0.05)。②经侧脑室给予150μg Pyrilamine预处理后,给予4nmol Orexin A溶液前后皮层神经元放电频率并无显著变化(n=15,P>0.05)。结论Orexin A对麻醉大鼠前额叶皮层神经元放电活动有明显的兴奋性,Pyrilamine对此兴奋性存在抑制效应。
Objective To study the indirect route and possible mechanism by which Orexin A regulates the cortex in the anaesthetized rats. Methods Fifty-five rats were divided into 4 groups,i, e. ①control(n=12) ,②Orexin A(n=16) ,③Pyrilamine(n= 11)and④Pyrilamine+ Orexin A(n=16) groups. The extraeelluar unit fuging of neurons in all the rats's prefrontal cortex were recorded by the glass microelectrode technique. Results ①The firing rate ,which come to the climax within 6 minutes after the administration of orexin A,significantly increased inx. the first 9 minutes after stopping injeceing the drug as compared with the basic discharging rate in Orexin A group ( P 〈0. 05 ). ②The increase in rate of prefrontal cortex neurons was insignificantly induced by 4 nmol orexin A solution after the pretreatment with intracerebrov- entficular injection of pyrihamine ( P〉0. 05). Conclusion Orexin A can exert a significant excitative effect on prefrontal cortex neurons in the anaesthetized rats, but the excitative effect can he inhibited by Pyrilamine.
