L-精氨酸制备大鼠急性轻症胰腺炎模型的研究 被引量:5

摘要 目的建立一种操作简单、病变典型、稳定性好的大鼠急性轻症胰腺炎模型。方法大鼠腹腔内分次剂量L-精氨酸注射(3×1g/kg)。注射后6、12、24、48、72h观察血清淀粉酶水平、胰腺病理学改变厦胰腺组织湿/干质量比值的变化。空白对照组以生理盐水代替,6h时处死。结果注射L-精氨酸6h后大鼠即出现典型的急性轻症胰腺炎病理改变,24h达高峰,48h出现少量胰腺细胞坏死,72h病变减轻;胰腺组织湿/干质量比值变化、病理评分呈时相性变化。血清淀粉酶6h时显著增高,24h达高峰,为对照组的3—4倍,72h接近正常水平。结论分次剂量L-精氨酸腹腔内注射诱导的大鼠胰腺炎症改变符合急性轻症胰腺炎病理学的改变,是值得推广的动物模型. Objective To establish a rat model with acute pancreatitis that is simple, stable and with significant metastasis. Methods Male rats were injected intraperitoneally with L-arginine (lg/kg) every hour within 3 hours. Control group rats were injected with the same quantity of 0.15 M NaC1. Rats were killed at 6, 12, 24, 48, 72h following L-arginine administration. The serum amylase level, wet/dry weight ratio of the pancreas and histologic scores were assessed. Results The serum level of amylase was significantly increase at 6h, reached the peak level at 24 h, and decreased to normal level at 72 h. Histologic examination revealed that interstitial edema and inflammatory cell infiltration was obviously observed at 24 h at the peak, Then histological scores decreased at 48h. The wet/dry weight ratio of the pancreas changed in accordance with the interstitial edema. Conclusion The study demonstrated that injection of L-arginine at multiple steps can induce noninvasive experimental model of mild acute pancreatitis.
